PCBAS-MAGNET-IL-18

PCBAS-MAGNETIL-18

Project integrated into the framework of PERTE for Vanguard Health and co-financed by the Spanish Ministry of Science, Innovation and Universities with funds from the European Union NextGenerationEU, from the Recovery, Transformation and Resilience Plan (PRTR-C17.I1) and from the Autonomous Community of Galicia within the framework of the Biotechnology Plan Applied to Health. The project MAGNET_IL-18

This project targets the development of magnetic nanostructures functionalized with cytokine IL-18, to induced a controlled inflammatory response in different types of cells.

Objectives

  1. Development of magnetic microstructures formed through the deposition or growth of spherical or elongated iron oxide nanoparticles onto polystyrene (PS) spherical substrates, in order to modulate the final size of the microstructure, surface topography, and magnetic response. Additionally, each magnetic microstructure will be individually coated with COFs, thereby increasing the available surface area for a more effective anchoring of the cytokine biomolecule IL-18. The anchoring of IL-18 will depend on the surface chemistry of the different magnetic microstructures employed, facilitating its subsequent release into the cellular environment. (Months 1–12).
  2. Evaluation of the internalization processes of cytokine-loaded microstructures in different cell types, either via endocytosis or magnetically forced internalization, leading to their entrapment in lysosomes or their release into the cellular cytoplasm. (Months 7–12).
  3. Assessment of the pro-inflammatory cellular response triggered by the presence of the cytokine IL-18 in lysosomes or in the cytoplasm. For this, cytokine-loaded microstructures will be incubated with endothelial cells and other cell lines. Subsequently, specific pro-inflammatory cytokines will be quantified, and the activation of the NLRP3 inflammasome pathway will be determined, with the aim of correlating clinical data from PWH with the results obtained from the cell cultures. (Months 13–18).
